Research Area:         Respiratory diseases are rapidly increasing world wide and chronic obstructive pulmonary disease (COPD), a common inflammatory lung disease in which people experience long-term inflammation of the lungs is a major cause of illness and death throughout the world. In fact COPD at current the fourth leading cause of death has no cure yet and is projected to be the third leading cause of death worldwide by 2020. Since chronic lung diseases in general are hardly diagnosed at their early stages, where the acute phase of inflammation could be pharmaceutically controlled, the pathological ineffective resolution phase of the inflammatory response represents an important target of treatment. In this context endogenous lipid mediators that modulate leukocyte trafficking and promote the inflammation resolution are seen as promising drug targets.
Therefore we aim to study the dynamics of pulmonary inflammation in mice, with a particular focus on the impact of pro-resolving lipid mediators. We hypothesize that secretolytic drugs including Ambroxol, known for their mucociliary clearance stimulating activity in the bronchial airways, have additional beneficial properties to overcome inflammation by producing anti-inflammatory and pro-resolving lipid mediators.
Ambroxol, known to inhibit pro-inflammatory mediators, has so far not been studied for its capability to induce pathways of anti-inflammatory and pro-resolving lipid mediator production such as prostaglandin E2, 15-hydroxytetraenoic acid or lipoxin A4, all being important members in the orchestra of mediators to terminate the inflammatory process.

Helmholtz – CSC – Fellowships 2010
Helmholtz Centre:        Helmholtz Zentrum München
Research Field:        Pulmonary Drug Delivery
Research Project:         Efficient delivery of aerosolized drugs into the lungs of mice
Position:        Postdoc

Department/Supervising scientist:  Institute of Lung Biology and Disease/ Dr. Otmar Schmid

Research Area:
Inhalation of macromolecules (e.g. proteins, peptides) is receiving increasing attention for pulmonary and systemic delivery of innovative drugs due to advances in genetic engineering and biotechnology. Numerous mouse models are available for proof-of-principle studies during the pre-clinical development of these therapeutic approaches.

In this project innovative techniques for efficient and dose-controlled delivery of aerosolized drugs into the lungs of mice are developed and validated in mouse models.

Specific Requirements:
-        Ph.D. degree in physics, engineering or a related field
-        Strong background in aerosol science
-        Interest in design and test of novel instrumentation
-        Willingness to work in a highly interdisciplinary field

Work Place:        Neuherberg/München

Earliest Start (between September 2010 and February 2011):  Sept. 2010

Research Project:         Determination of the biologically effective dose of drugs delivered to the lungs of mice

One of the most crucial parameters is not only knowledge of the dose delivery to the lungs, but more importantly, knowledge of the biologically effective dose delivered to the lungs.
In this project measurement techniques for determination of the biologically effective dose of drugs delivered to the lungs of mice are to be developed and validated.

Specific Requirements:
-        PhD in biology, pharmacology, medicine, veterinarian medicine or a related area
-        Strong background in genetics as well as cellular and molecular biology
-        Experience with disease models in the mouse
